01Project Introduction

02Excellent performance of the fluorescent cell counter in the project
This project involves a large amount of cell counting work, and since the counted cells are from peripheral blood, the components in the samples are complex, especially with possible contamination from red blood cells, which are similar in size toPBMC, this makes traditional counting methods unable to meet technical requirements.
PBMCAfter mixing the sample with fluorescent dye, the nuclei of individual nucleated cells are stained, while red blood cells cannot be stained due to being anucleate, making it perfectly suitable for the counting work of this project.

The cell counter used in this project comes fromNewton OpticsJupitaFluorescent Cell Analyzer.The following is the instrument interface, which cansimultaneously display the counting results of multiple samples, including quantity, viability, average cell diameter, aggregation rateand so on.

The following shows the difference between bright field counting and fluorescent counting of samples. It can be seen that due to the presence of platelets and red blood cells in the bright field, the view is very messy, and the counting is inaccurate. Under fluorescence, nucleated cells are clearly displayed, and the counting is accurate and clear.

The following is a comparison of counting data between the fluorescent counter and flow cytometer, which shows that the counting results are not significantly different. (Due to sampling, mixing, instrument, and other errors, complete elimination is not possible)

IPSCell quality standards:
1.DetectionhiPSCs' pluripotency markersOct4,Sox2,TRA-1-81,NanogStem cell marker genes are stably expressed in more than90%of the cells.
2.Karyotype testing is qualified, excluding abnormal chromosome recombination.
3.Provide short tandem repeat(short tandem repeat,STR)detection report, establishing a uniqueDNAidentity for each sample.
4.Provide detection reports, with no fungal, bacterial, or mycoplasma contamination.
