01 Laboratory Introduction

Hubei Hongshan Laboratory was 2021 year 2 month, officially established and approved by the People's Government of Hubei Province. The laboratory focuses on the field of bio-breeding and currently brings together a group of outstanding scientific and technological talents, including 7 national / National Key Laboratories, 7 National Natural Science Foundation of China Innovation Research Groups, 8 academicians, including a number of Changjiang Scholars, National Outstanding Young Scientists, and other outstanding scientific and technological talents. 02 Experimental Background

Cell Passage: Cell passage is a key operation in cell culture, aimed at allowing cells to continuously grow and proliferate, providing a sufficient cell source for scientific research, etc. When cells in a culture bottle grow to 80% - 90% confluence, they need to be passaged.

 

03 Experimental Steps

1. Observe cell status: Observe the cells in the culture bottle under a microscope. When the cell confluence reaches 80% - 90% ,the cells are plump, growing vigorously, and show no obvious signs of contamination, it is suitable for passage.

2. Preparatory Work: Place the required experimental equipment in the ultra-clean workbench and wipe it with 75% alcohol for disinfection. Turn on the ultraviolet lamp of the ultra-clean workbench and irradiate it for 30  minutes for sterilization. At the same time, place the culture medium, PBS ,trypsin, etc. into Newton Optics Cell Recovery Instrument in " Waterless heating ,set 37 degree preheating.

3. Discard the old medium: In the ultra-clean workbench, open the lid of the culture bottle and carefully aspirate the old medium in the culture bottle with a pipette or pipette, trying to aspirate as much as possible.

4. Wash cells: Add an appropriate amount of PBS , gently shake the culture bottle to PBS thoroughly rinse the cell surface, then aspirate and discard. Repeat this step PBS times to remove residual serum and metabolic waste from the cell surface. 1 - 2 次,以去除细胞表面残留的血清和代谢废物。

5. Digest cells: Add an appropriate amount of trypsin solution to the culture bottle, just enough to cover the cell layer. Place the culture bottle in 37℃ of CO incubator for incubation. The digestion time depends on the cell type, generally 1 - 5 minutes. During this period, the cell status can be observed under a microscope. When the cells become rounded, the gaps between cells increase, and some cells begin to detach, it indicates that the digestion is moderate.

6. Terminate digestion: Quickly add an appropriate amount of complete medium containing serum to the culture bottle to terminate the trypsin digestion. The proteins in the serum can bind to trypsin, rendering it inactive.

7. Blow cells: Gently blow the cell layer with a pipette or pipette to fully disperse the cells into a single cell suspension. When blowing, pay attention to gentle movements to avoid producing a large number of bubbles, which may damage the cells.

8. Cell counting: Take a small amount of cell suspension and use Newton Optics Cell Counter to calculate the cell concentration and viability to determine the number of cells to be passaged and the inoculation density.

9. Subpackage cells: According to experimental requirements and the size of the culture vessel, use a pipette to subpackage an appropriate amount of cell suspension into new culture bottles or dishes. Then add an appropriate amount of complete culture medium to each culture vessel, gently shake it to evenly distribute the cells.

10. Culture cells: Place the culture bottles or dishes with subpackaged cells in 37℃ 5% CO incubator for culture. The temperature, humidity, and gas environment in the incubator can provide suitable growth conditions for the cells.

04 High-efficiency and precise cell analysis solution
