Experimental Background

Gene sequencing technology, which determines the sequence of DNA bases to decode the genetic information they carry, has become a cornerstone tool in modern life sciences. Single-cell sequencing, as an important branch of gene sequencing, enables high-throughput sequencing analysis of genomic, transcriptomic, and epigenomic information at the single-cell level. It can reveal the genetic structure and gene expression status of individual cells, reflecting cellular heterogeneity and playing a crucial role in fields such as oncology, developmental biology, microbiology, and neuroscience.

 

 

Highly efficient and precise cell analysis solutions

 
 

 

 

In single-cell sequencing, the core role of the cell counter lies in its function as a critical initial quality-control step. By providing precise quantification and analysis, it ensures the quality of upstream samples and directly determines the reliability of downstream sequencing data. Using dual-fluorescence staining techniques (such as AO/PI), this method specifically identifies nucleated cells and accurately measures cell concentration, viability, and diameter, effectively eliminating interference from red blood cells, platelets, and cellular debris. Moreover, it can distinguish between intact cells and isolated nuclei—for example, the average diameter of live cells is 13.4 μm, whereas the average diameter of fully separated nuclei is 8.3 μm. Ensure that the samples meet the stringent requirements of single-cell sequencing platforms (e.g., total cell count > 5×10⁵, viability > 70%, clumping rate < 10%). Thus, it ensures from the source the efficiency of single-cell capture as well as the accuracy of subsequent sequencing data.
